Charlie Rose: May 21, 1993 This is an episode of Charlie Rose. It is an AIDS discussion. Laurie Garrett is a journalist who documents medical news. She wrote The Coming Plague: Newly Emerging Diseases in a World out of Balance. It was published in 1994 and covers developments on HTLV-I & II and the HIV (or HTLV-III ), the spread of AIDS into remote populations, etc. The other speakers are equally as good. You probably saw things like this 13 years ago. It is a good review. The people who are most effective at fighting AIDS are themselves it seems. Unfortunately the modes and methodologies is expensive. So expensive that almost all research seem…

I need help in conducting a cross examination of HCV associated Non-Hodgkins Lymphoma and HCV associated Lymphoplasmacytic Lymphoma. As far as my research has taken me I have discoverd that for an unknown reason HCV associated NHL has a higher response to Anti-Viral Treatment (Interferon) than HCV associated LPL. Thus far the two points of commonality I have established are the HCV protein and the rearangement of of the PAX-5 Gene as is common among plasma diffuse lymphomas. My knowledge of the PAX-5 gene is limited to it's function of encoding B-Cell specific Activator Protein (S). I am attempting to use this line of inquiry as an attempt to isolate the key factor which…

I am an undergraduate working on a final project and I need some help if anyone can... I have run FACS analysis of thymocytes from 508 and 506 TCR tsg Rag -/- mice exposed to the WT 3K peptide and 6 alanine subsitutions (mutated variants) and have found evidence of negative selection occuring (death correlating with the increase in CD69 expression). I also have data from harvested splenocytes (proliferation assay) that demonstrate positive selection correlating with the increase in CD69 expression. These two things pretty much just demonstrate that the peptide-MHC-TCR conjuction is effective in allowing development to proceed. However I am supposed to identify…

Bird flu H5N1risk as a significant pandemic - despite all the scares and hype - appears to be fading as a possibility. WHO stats for 2007 show a clear decline and total deaths in all the years since description approximate only 200 world wide. Is this yet another screw up by WHO/CDC et al.? Will bird flu H5N1 join BSE, SARS, swine flu even Ebola in the pandemic public health scare "never mind" circular file?

Hey guys, Anyone following the Dover case could see Dr Behe's rediculous approach to the dozens of books explaining the evolution of the immune system through natural selection. I find the subject of evolution in general fascinating, but most of the resources I found online about the evolution of the Immune system were *waaaaay* beyond my level. I am not sure if I was just unable to find any good 'lay men' resources or if it's just a dificult subject, but I was wondering if anyone could perhaps give me a brief layman explanation on how the immune system evolved.. where does it come from? Or.. well.. resources would be fine too. Thanks! ~moo

Hiya, I am new to this forum so feel a bit cheeky asking for help first off but my brain is in totally blonde mode. I am a third year degree student and my project is in novel antimibrobial products... I plan to look at levels of inhibition at depth thus simulating wound depth but my mental block is with the methodology. I plan to puncture agar at different depths, inocculate these depths with MRSA and then apply the antimicrobial and asses its properties from this, but i cant think of a standard peice of equipment from which to do this as i need something deep like a beaker but it needs to be able to be sealed and the flatter the better for visable colony counting... i…

Hi all, I was wondering if anyone could help me regarding a microbiology assignment. I am confused and need some guidance as to what to do next. Details of the assignment below: Q) Describe, how food poisoning bacteria are able to survive and proliferate in foods. Taking in to account the following factors: •The influential factors within foods which influence the growth and survival of microorganisms. • Relate these factors to the growth of specific food borne bacterial pathogens •Use information from journals (at least five references published within four years) to Effectively explain the relationship of environmental factors and the growth of s…

Hello all. I was wondering how much (what concentration) of LPS per ml of cell-medium, do you have to use to keep the PBMC culture alive and proliferating. The PBMC is from a healthy donor. Thanks in advance.

Thinking out aloud here. What makes a strong immune system and one to be healthy, no physical / chronic health conditions at all? I find it fascination really. Some people can exercise religiously, eat a healthy and balanced diet, not smoke, not drink alcohol and seem to 'catch' anything and everything. Yet there are others who don't look after themselves and never get sick. See, I am one of them. It's very odd, but lucky I guess. I smoke, have done for 7 years, my diet mainly consists of toast, pasta, dry biscuits and coffee - very rarely eat fruit or vegetables, don't eat meat, cheese or eggs. It isn't genetics. My parents are always sick with gastro bu…

hi, I incubate cells for 2h in 37°C 5%CO2 and try with a cell scraper to scrap my cells which adhered on the petri-dish. I want to distinguish between adhere and non-adhere cells. Can anyone tell me how is the best way to do it because Im loosing cells (I count the cells in supernatant and cells which adhere on petri-dish. The total amount is quite far away from my original input). thx, Annie

Hello, I am using the ELISA immunoassay for allergen testing. What is the proper way to manually wash the microtiter plates with PBS-T? Is it ok fill the wells with PBS-T with a squirt bottle (not overflowing), and then quickly invert the plate and tap it? Or will doing it this way cause cross-contamination between the wells? Any help would be greatfully appreciated.

Hi! I have a couple of questions regarding the induction of lasting immunity by vaccination: 1) To induce BOTH a cellular and humoral response by vaccination, is it sufficient for antigen presenting cells to endocytose, digest and display peptides in the MHC-II complex? This would present antigens to CD4+ helper cells, but I am not sure whether the following cascade would be enough to induce a strong response both from cytotoxic T cells and antibody secreting B cells.. The alternative, I suppose, would be to have a vaccine that also entered the cytoplasm of cells, thereby gaining access to the MHC-I complex, which is recognized by CD8+ cytotoxic T cells. Any t…

Hello, I have a question regarding regulatory experiments using generated regulatory T cells and adding them to different DC:T (responder) cells allogeneic cultures. The problem is that by adding Tregs, you also dilute the existing DC:T(resp.)cell ratio and thereby automatically reducing cpm counts. Does anyone have an idea how to cope with this, becasue in the literature they don't seem to have a problem with it, but I do. When they put control effector cells instead of Tregs, it doesnt lower their cpm count, although the number of effector T cells is as high or higher than that of responder T cells. Thank you very much, best regards.

HIV is a serious threat to present and future generations, as you will know if you follow the news and science papers. It would be interesting if we could maintain a 'Think Tank' to collate current approaches to fighting the virus, and to process the plausability of conjectural treatment. It has occurred to me recently that where there has been much research into the prevention of T4 - HIV interactions through inhibitors etc, there does not seem to be any research that exploits the virus' affinity for this structure. So, some questions: Red blood cells have a far greater volume than T4-lymphocytes, no nucleus, and are more numerous by a long way. Would…

Hello I have been following the ongoing worldwide study of influenza pandemics of the past, H5N1 with interest. I have a question regarding virusses in general. My Question I have heard genetic scientists talk about virusses and at this point in history we seem to know quite a lot about their genetics etc.. When it comes to their origin however, as far as I know we don't know how virusses came into being. I have heard ideas that they may be remnants of primordial dna or nucleic acid systems but this answer doesn't completely satisfy my curiosity. It seems to me that a virus is well equipped to invade the cells in many different types of body tissu…
