I'll be doing some molecular cloning this year. Any advice on protocols and data analysis?

Does it provide raw data that are analysed? It's much easier to understand concepts when you can work through them.

Cloning with plasmid vectors is pretty straightforward. Follow the kit instructions and pay close attention to sterile technique and you should get beautiful sequence as a result of the M13 primers most of them use.

Yeah, there is not a lot in terms of raw data (compared to other analytical techniques) that I could think of. The closest is probably size determination. Once one does certain quantitative methods, such as QPCR the data is still fairly simple, but normalization tends to be the challenge.

 

Once you understand what the steps are, it is really a procedure which could be performed while being asleep (or being really drunk).

I haven't started with any of cloning yet. So, I'm unsure what you guys are talking about.

 

I'm still doing centrifugation, SDS-page and chromatography.
