side of the combs in the tray for the agarose gel

Hi all,

 

I have a very stupid question.Is it important the side of the combs when you place them in the tray for the agarose gel to form the wells before pouring the gel into the tray? Can you explain me why?

Pour the gel into the tray, then place the comb in the gel..remove the comb. That's how I've always done it. I think its just better to pour gel, then comb instead of the other way around..I guess just an industry standard.

Unless they have specialized notches either way works fine. Putting them in first helps to avoid pushing in bubbles sometimes, but really is a matter of preference.
