This is a sample question. I have a test tomorrow and I am so lost with this stuff. help me out pls!

 

2) The placement of the Z-ring during cell division is mediated by the Min system and Nucleoid Occlusion. While knocking out the Min system causes a significant amount of polar divisions, nearly normal growth is observed. However, when min- is paired with other knockouts; such as slmA- (involved in nucleoid occlusion) or rodA- (involved in cell shape), cell growth is seriously affected. Describe a screen that may allow you to discover other novel factors involved in cell division. After finding a mutant, how will you characterize the role the gene plays in cell division?

 

So far I have (btw this might not be correct so please correct me anywhere):

 

min abnormality or knockout by itself doesn't have a lethal outcome. However, when it is knocked out or mutant in combination with another factor such as slmA, the cell dies. So in order to find other factors like SlmA, I would make a mutant (random mutations) library of an already knocked out min system with defective dna polymerase or transposons? I would screen for cells that have been affected in some way to a degree, and isolate them. Now what?? How do i find out where the mutation took place in say, one defective cell that I isolated. and lastly, how do i characterze the role this gene plays in cell division? GFP localization, etc? so lost.
