Hello everyone, this is my first time to post a topic over here, anyways, I'm a master student and I'm supposed to do Astrocyte primary culture from mouse cortex (mice p2-p5), I've tried it today for the first time and I ruined off the experiment, I guess all the cells that I got were dead already or they died from incubation in trypsin, I'm not actually sure. I do have several protocols for that but I wish if someone is using a specific protocol that is good and works to inform me about it and please from the very beginning as I still have problems with separating the cortices from the pups. Thank you.
