Migration assay using protein extracts?

I would like to know if someone has ever done migration assay using tissue or cell protein lysate (instead of conditioned media or media + chemoattractant)?

If so, is there a better protein extraction protocol that would keep proteins stable for at least 24 h at 37 degrees?
