Isolating a gene from specific allele

is it possible to isolate  a specific gene from specific allele?

We are trying gene suppression research and we know that one gene has a negatively affecting mutation. While we can suppress the mutant gene via allele specificity - we aren’t certain if a good gene in another allele functions as expected.

 

Hence we are curious how to isolate the functional gene from another allele.

Any help and details are appreciated
