Is it possible to calculate de novo mutation rate in a genetic disease ?

Hi All, 
I am wondering if it is possible to calculate the de novo mutation rate in a genetic disease, given that you know the current disease incidence rate ? Does the de novo mutation rate change over time, as a result of environmental factors etc ? I'm new to genetics so I'm not really sure what plays a part in de novo mutation rates. I can image that if there was an equation, it would take into account many parameters.
