Anyone out there with any experience? I'd be grateful for a link to a good trouble-shooting guide. I've been embarked on this for a few weeks but never having done it before I'm not too aware of the pitfalls. Any good websites gratefully recieved!!

What exactly are you doing in relation to protein expression in bacteria...?

I've moved on a bit now....any tips on site directed mutagenesis?? I can't get it to work. I've tried changing template amounts, DMSO, used a gradient block to vary annealing temp, used a different clone, tried a kit, tried the standard lab protocol.....:confused: Any magic wands out there?
