Does anyone work with or understand PAM fluorometry for the purposes of assessing ETR rate in microalgae? We have a Phyto-PAM, but not sure how to operate - everything is set up, but how to use the software to get the correct results is a bit shaky. I think if I understood what results I am supposed to be getting, then I will be able to play around with the settings - but I am completely new to this. And no one else at work seems to be able to help me either... I've attached the manual of the PAM we use. Thank you! phyto_4e.pdf

Have the results been 'reproduced' before - more than one reference? If you are doing the same thing as was done before (exactly), you should be getting the same results. Otherwise, there has to be something wrong with the compound. You are using the same concentrations and everything? -

It's likely that there is something wrong with your test compound, otherwise is it absolutely certain that this particular compound causes expression of the enzyme? -

Yes - your arguments has givn me much insight regarding this matter. I would however like to discuss it some more as there are still a few points I would like you to clarify for me.

It was good to have seen you today Monique! I particularly enjoyed our in depth discussion on certain genetic disorders that seem to have become more prevalent. Or perhaps it is our awareness that have become increasingly sensitive.
