iRNAblogger Posted August 27, 2014 Share Posted August 27, 2014 Are you concerned with the typology of the plasmid? (Such as amount of supercoiled vs relaxed vs nicked?) Because some studies have suggested that temperature changes can affect the supercoiled state of plasmids: http://www.pnas.org/content/81/13/4046.full.pdf+html and I'm sure that a similar phenomenon could be observed due to freeze-thaw cycles.
