  1. You mean three different concentrations of different proteins, or three different concentrations of the same protein? You added Ellman's reagent (DTNB) and citrate synthase (your protein?), and measured the absorbance of the formation of the product of the reaction (TNB) at 412 nm?
  2. Glucose --> glucose 6 phosphate = transfer of γ-phosphate of ATP to glucose, using the energy from hydrolysis of γ-phosphate of ATP Glucose 6 phosphate --> fructose 6 phosphate = isomerase reaction = net energy change is 0 kJ.mol^-1 Fructose 6 phsophate --> fructose 1, 6 bisphosphate = same principal as first reaction (transfer of a γ-phosphate from ATP, using the energy of hydrolysis of the γ-phosphate)
  3. Hi, Would someone be able to confirm or correct my understanding of cPKC regulation? cPKCs bind DAG by their C1 binding domain, and bind Ca2+ by their C2 binding domain, and then (this is this bit I'm slightly unsure of), the Ca2+ bound C2 domain can then bind to phosphatidylserine. When all of the aforementioned interactions are satisfied, cPKCs are fully active? Thanks
